Research Areas

LSA Technology Development Group implements the development, introduction, and standardization of the technologies, especially those for next generation sequencers, which constitute the Life Science Accelerator (LSA). The group maintains a close connection with the LSA System Development Group, Functional Genomics Technology Team, and the LSA Bioinformatics Core Facility in order to contribute to the leading research of the Omics Science Center. LSA technologies include exploration of novel pathways in gene expression regulation, development of various research tools by utilizing genomics resources, technology development for detecting genome variation, technology development for analysis of intra-gene interaction, development of various molecular libraries subjected to the LSA, and systematic proteome analysis.
A special task of this group is to achieve large-scale operations, at the level of the FANTOM (Functional ANnoTation Of Mammalian genome) projects, where technologies are scaled up in throughput and broadly tested on some yet unsolved biological problem. The FANTOM international meetings have been held five times and produced fruitful results as follows:
FANTOM1-3: The consortium developed an effective system for functional gene annotation (FANTOM1), and analyzed more than 60,770 full-length mouse cDNAs, which was published in a special issue of Nature on the decoding of the mouse genome (FANTOM2). In the FANTOM3, we published a couple of papers in the "RNA special issue" of Science in 2005, where we reported comparable existence of long non-coding RNA as well as protein coding RNA as "Discovery of new RNA Continent".
FANTOM4: We aimed to elucidate the transcriptional regulatory networks to understand biological phenomena at the molecular level. The development of the network analysis methods and findings of novel RNAs were published in three papers in the 'Nature genetics' special issue. Other results were published in the FANTOM special issue of 'Genome Biology', and results of the protein-protein interaction analyses of transcription factors in 'Cell'.
FANTOM5: We succeeded to develop high quantitative CAGE technology for application to the next generation sequencers. Based on this technology, promoter-centered large-scale transcriptome analyses are going on for various cell types in human and mouse.
Furthermore, as the application of above mentioned transcriptional regulation analysis, we developed a novel factor to promote pluripotency in mouse iPS cells, and direct cell conversion technology from human fibroblast cells.
